M1” macrophages were generated in vitro from human primary CD14 + monocytes as described above. “M1” macrophages were plated into an opaque-walled 96-well plate at 10 × 10 5 cells per well and incubated with a concentration gradient of GSK761 (0.04–1.11 μM) for 1 h (0.1% DMSO was used as control). An equal volume of freshly prepared CellTiter-Glo® reagent was added to each well, the plate was shaken for 10 min at RT, and luminescent signals were recorded using a plate reader (SpectraMax M5). The index of cellular viability was calculated as the fold change of luminescence with respect to untreated control cells.
