For the mutant assay, the labeled mutant probe was added to the reaction system for 40 min. In the antibody-based assay, the labeled probe, nucleoprotein, and 1 μl of Flag antibody were incubated at 25 °C for 40 min. The DNA-protein complex was separated on a 5% nondenaturing polyacrylamide gel in 0.5x TBE buffer (Beyotime Biotechnology, Shanghai, China) by electrophoresis at 100 V for 80 min. Finally, images were obtained with a Tanon-5500 Chemiluminescent Imaging System (Tanon, Shanghai, China).
